RSPO1‐Accelerated CEC Proliferation and Inhibited MT Through the Wnt Pathway. Previously, LGR5 was thought to be an orphan receptor of the G protein‐coupled receptor superfamily, and its ligand was unknown. However, several recent reports demonstrated that RSPOs function as ligands of LGR5 to regulate Wnt/β‐catenin signaling [35-37]. LGR5 was the top gene up-regulated in ZG (25-fold). The gene for its cognate ligand R-spondin-3, RSPO3, was 5-fold up-regulated.In total, 18 genes associated with the Wnt pathway were greater than 2-fold up-regulated.

Our genetic experiments in leukemia models demonstrated that LGR5 is critical for leukemia-initiation and survival. Abstract. Lgr5 was originally discovered as a common Wnt target gene in adult intestinal crypts and colon cancer. It was subsequently identified as an exquisite marker of multiple Wnt-driven adult stem cell types. Lgr5 and its homologs, Lgr4 and Lgr6, constitute the receptors for R-spondins, potent Wnt signal enhancers and stem cell growth factors.

Lgr5 expression is Myb-dependent.

Its specific roles during inner ear development are just emerging, but few studies have characterized Wnt target genes. Lgr5, a member of the G protein-coupled receptor family, is a Wnt target in the gastrointestinal and integumentary systems.

Lgr5/Gpr49 is a Wnt target gene as well as a cancer gene; it was on the original list of Wnt/Tcf4 targets active in colorectal cancers (van de Wetering et al, 2002) and is overexpressed in tumours of the ovary and liver, likely because of mutational activation of the Wnt pathway in these tumours (Yamamoto et al, 2003; McClanahan et al, 2006; Zucman‐Rossi et al, 2007).

However, gain-of-function studies using RSPO ligands and a new non-lipidated Wnt analogue reveal that these ligands have qualitatively distinct, non-interchangeable roles in ISCs. 2011-07-04 · Lgr5 homologues are facultative Wnt receptor components that mediate Wnt signal enhancement by soluble R-spondin proteins. These results will guide future studies towards the application of R Lgr5 was originally discovered as a common Wnt target gene in adult intestinal crypts and colon cancer. Recently, leucine-rich repeat-containing G-protein-coupled receptor 5 (LGR5) is a newly identified cancer stem cell marker and Wnt target gene. However, the role of LGR5 in gastric cancer (GC) remains uncertain. This study was performed to investigate the effect of LGR5 expression in GC. 2020-04-15 · Because LGR5 is a target of Wnt/β-catenin signaling that occurs during the adenoma–carcinoma sequence in CRC, LGR5 expression in MMR-P cases, which may involve the adenoma–carcinoma sequence, may be higher than that in MMR-D cases, which may involve the serrated neoplasia pathway . 以前より腸幹細胞は、Wnt signalにより増殖が調節されていると報告されていました。そこで著者らは先行論文でWnt signalのターゲット遺伝子であるLgr5に着目しました。 The receptor LGR4 promotes Wnt signaling in response to the binding of R-spondin ligands (RSPOs) both by inhibiting the activities of the E3 ligases RNF43 and ZNRF3 and by directly stimulating the Wnt signalosome through the scaffold protein IQGAP1. It is thought that LGR5 stimulates Wnt signaling similarly to LGR4 although LGR4 and LGR5 are not functionally equivalent in vivo.

Lgr5 is thought to function as a potentiator of Wnt/β-catenin signaling in epithelial cells. Upon deletion of Lgr5 in B-ALL cells, however, we observed massive accumulation of nuclear β-catenin and increased expression of β-catenin target genes, which suggests that Lgr5 acts as an essential negative regulator of β-catenin in the B-cell context. Wnt signaling regulates Lgr5 expression.
